Antibody–drug conjugates (ADCs) have emerged as one of the most sophisticated therapeutic modalities in modern biologics manufacturing.

By combining the targeting precision of monoclonal antibodies with highly potent cytotoxic payloads, ADCs enable the direct delivery of treatments to diseased cells while limiting systemic exposure.

The growing importance of cleaning validation in ADC biologics manufacturing

This therapeutic promise has accelerated investment in ADC pipelines across the pharmaceutical industry; in the US alone, 430 ADC studies were actively being conducted in 2024.1

But the complexity of ADC manufacturing has also introduced new operational and regulatory challenges, particularly regarding payload containment and cleaning validation.

Unlike conventional biologics, ADCs involve the handling of highly potent linker–payload compounds that must be carefully controlled throughout development and production.

Even trace residual levels can compromise product quality and increase the risk of cross-contamination in manufacturing environments that use hybrid and traditional stainless-steel equipment.

As ADC pipelines continue to expand, manufacturers are under increasing pressure to establish lean yet highly sensitive validation strategies that support operational efficiency without compromising safety.
